TIP-15 was previously identified as a cellular protein that can bind to the C-terminal end of the HTLV-1 Tax protein via its two PDZ domains. The sequence of the N-terminal part of TIP-15 is identical to that of the synaptic protein PSD-95. Both proteins are likely to be produced from the same gene by alternative splicing. Whereas expression of the PSD-95 mRNA was detected only with brain RNAs, that of TIP-15 was detected with RNAs from thymus, brain, skeletal muscle and Jurkat cells. The TIP-15 protein exhibits an apparent molecular weight of 40 kD and is weakly expressed in T cell lines. A two-hybrid screen performed with TIP-15 as bait revealed the presence of a PDZ binding site (PDZ-BS) in the following proteins: Lysyl tRNA synthetase, 6-phosphogluconolactonase (6-GPL), Stress-activated protein kinase 3 (SAPK3), NET-1, Diacylglycerol kinase zeta, MTMR1, MCM7, and hSec8. The sequence at the C-terminal ends of these proteins matches the X-S/T-X-V-COOH consensus previously defined for PDZ-BSs, with the exception of 6-GPL and SAPK3 which include a leucine as the C-terminal residue. For Lysyl tRNA synthetase, NET1, MTMR1 and hSec8, binding to TIP-15 was confirmed by co-immunoprecipitation experiments performed with the extracts of transfected COS7 cells. These results show the existence of functional PDZ-BSs in these proteins, but future studies will be necessary to establish whether or not TIP-15 represents a physiological partner. The significance of the presence of a PDZ-BS in these various proteins is discussed with respect to their function. 相似文献

Abstract: Cultured cerebellar granule neurons undergo apoptosis when switched from a medium containing depolarizing levels of K+ (25 mM KCI) to medium containing lower levels of K+ (5 mM KCI). We used this paradigm to investigate the role of caspases in the death process. Two broad-spectrum caspase inhibitors, tert-butoxycarbonyl-Asp·(O-methyl)·fluoromethyl ketone and benzyloxycarbonyl-Val-Ala-Asp·fluoromethyl ketone, significantly reduced cell death (90 and 60%, respectively) at relatively low concentrations (10–25 µM), suggesting that caspase activation is involved in the apoptotic process. DNA fragmentation, a hallmark of apoptosis, was also reduced by these caspase inhibitors, suggesting that caspase activation occurred upstream of DNA cleavage in the sequence of events leading to cell death. As a step toward identifying the caspase(s) involved, the effects of N-acetyl Tyr-Val-Ala-Asp·chloromethyl ketone (YVAD·cmk), an interleukin-1β converting enzyme-preferring inhibitor, and N-acetyl Asp-Glu-Val-Asp·fluoromethyl ketone (DEVD·fmk), a CPP32-preferring inhibitor, were also evaluated. YVAD·cmk provided only modest (<20%) protection and only at the highest concentration (100 µM) tested, suggesting that interleukin-1β converting enzyme and/or closely related caspases were not involved. In comparison, DEVD·fmk inhibited cell death by up to 50%. Western blot analyses, however, failed to detect an increase in processing/activation of CPP32 or in the proteolysis of a CPP32 substrate, poly(ADP-ribose) polymerase, during the induction of apoptosis in granule neurons. Similarly, the levels of Nedd2, a caspase that is highly expressed in the brain and that is partially inhibited by DEVD·fmk, also remained unaffected in apoptotic neurons undergoing apoptosis. These results suggest that a DEVD-sensitive caspase other than CPP32 or Nedd2 mediates the induction of apoptosis in K+-deprived granule neurons. 相似文献

The cell cycle of donor cells as a major factor that affects cloning efficiency remains debatable. G2/M phase cells as a donor can successfully produce cloned animals, but a minimal amount is known regarding nuclear remodeling events. In this study, porcine fetal fibroblasts (PFFs) were carefully synchronized at G1 or M phase as donor cells. Most of the cloned embryos reconstructed from PFFs at G1 (G1-embryos) or M (M-embryos) phase formed a pronucleus-like nucleus (PN) within 6-h post fusion (hpf), but the M-embryos formed PN earlier than the G1-embryos did. Moreover, 77.4% of the M-embryos formed two PNs, whereas the G1-embryos formed a single PN. The rate of extrusion of polar body-like structures by the M-embryos was significantly lower than that extruded by the G1-embryos (26.3% vs. 37.1%, P?0.05), and DNA synthesis in most embryos in both groups was initiated at 9–12 hpf. Most of the M-embryos were octoploid before the first cleavage. Furthermore, 81.25% of the blastomeres of blastocysts developed from the M-embryos showed abnormal ploidy compared with those developed from the G1-embryos (22.55%). However, some of the blastomeres remained diploid in all the M-embryos tested. A portion of the blastomeres restored normal diploidy in some of the M-embryos at the blastocyst stage. It is generally assumed in dendroecological studies that annual tree-ring growth is adequately determined by a linear function of local or regional precipitation and temperature with a set of coefficients that are temporally invariant. However, various researchers have maintained that tree-ring records are the result of multivariate, often nonlinear biological and physical processes. To describe critical processes linking climate variables with tree-ring formation, the process-based tree-ring Vaganov–Shashkin model (VS-model) was successfully used. However, the VS-model is a complex tool requiring a considerable number of model parameters that should be re-estimated for each forest stand. Here we present a new visual approach of process-based tree-ring model parameterization (the so-called VS-oscilloscope) which allows the simulation of tree-ring growth and can be easily used by researchers and students. The VS-oscilloscope was tested on tree-ring data for two species (Larix gmeliniiand Picea obovata) growing in the permafrost zone of Central Siberia. The parameterization of the VS-model provided highly significant positive correlations (p < 0.0001) between simulated growth curves and original tree-ring chronologies for the period 1950–2009. The model outputs have shown differences in seasonal tree-ring growth between species that were well supported by the field observations. To better understand seasonal tree-ring growth and to verify the VS-model findings, a multi-year natural field study is needed, including seasonal observation of the thermo-hydrological regime of the soil, duration and rate of tracheid development, as well as measurements of their anatomical features. 相似文献

Fast protein liquid chromatography on Superose 6 of crude extracts from Chlorella kessleri, Fott et Novákóva, grown autotrophically in blue or in red light yields three different oligomeric forms of phosphofructokinase (PFK, EC 2.7.1.11). Their substrate affinities and responses to homotropic and heterotropic effectors are different. In vitro, the degree of oligomerization of the enzyme can be influenced by specific intermediates or cofactors. Its substrate, MgATP (10 mM/5 mM), and the negative effector, phosphoenolpyruvate (5 mM), both lead to some dissociation, while the second substrate, fructose-6-phosphate (5 mM), and the positive effector, inorganic phosphate (50 mM), have no effect. It is discussed whether formation or dissociation of oligomeric PFK forms in vivo result from alterations in the levels or in the intracellular distribution of effector molecules and whether such processes are involved in the different regulation of cell metabolism in blue or in red light. 相似文献
